Why DNA Fragmentation Matters in Male Fertility
Conventional semen analysis (count, motility, morphology) looks at how many sperm there are and how they move or look. But DNA fragmentation tests examine whether the DNA inside sperm is intact and undamaged.
High levels of sperm DNA fragmentation (SDF) are linked to:
- Reduced natural conception rates
- Lower success in IUI, IVF, or ICSI
- Higher risk of embryo arrest or miscarriage
When is testing especially useful?
In practice, DNA fragmentation testing is especially useful in cases of:
- Unexplained infertility (normal semen parameters but no pregnancy)
- Recurrent miscarriage (female factors ruled out)
- Poor outcome in prior IVF/ICSI cycles
- Lifestyle or exposure risk (smoking, toxins, heat)
MDC performs SCD testing in our in-house lab, delivering precise results you (or your fertility specialist) can use to guide decisions.
What the Test Measures & How
We use validated methods (such as TUNEL, SCSA, Comet, or SCD) to detect DNA strand breaks, nicks, or chromatin integrity issues in sperm. The test yields a DNA Fragmentation Index (DFI) or similar percentage metric.
Typical Thresholds (DFI)
- Low / Normal: usually < 20–25%
- Elevated: ~25–30% or higher — may suggest increased risk to fertility outcomes
Because methods and cut-offs vary by lab, interpretation always factors in the method used, your overall fertility profile, and clinical context.

Frequently Asked Questions (FAQs)
Does the test guarantee answers?
No. DNA fragmentation testing is a tool. It adds depth to your fertility profile, but it does not determine everything. Results must be contextualised by your clinician.
Is it painful or invasive?
No - it uses the same semen sample as standard analysis. There’s no extra discomfort.
How reliable is the test?
It depends on lab quality, sample handling, and method. That’s why we partner with accredited labs and follow strict protocols.
If I have high fragmentation, what next?
Your fertility specialist may recommend:
- Lifestyle and antioxidant therapy (if oxidative stress suspected)
- Sperm selection techniques (e.g. ICSI with advanced selection)
- Use of donor sperm (in extreme cases)
- Repeat testing after intervention
How often should I repeat it?
If you change lifestyle / receive interventions, repeating after ~3–6 months (one or two sperm cycles) is reasonable.
Does MDC treat fertility?
No - we are a diagnostic centre. Our role is to supply high-quality data (reports, lab values) to assist your doctor or fertility clinic.
